The in vitro and in vivo studies showed that the interaction of IGF-1 and IGF-1R is essential for cell growth and survival of various transformed cells [94]. Through furin and/or PC5, the IGF-1R, a heterotetramer of disulfide-bound α2/β2 chains, can be produced from endoproteolytic processing of a larger pro-IGF-1R at the RKRR740↓motif [95].

Insulin signal is initiated by insulin binding to the receptor and then activating the tyrosine protein kinase of the intracellular domain of the insulin receptor β-subunit (Baserga, 1999). The insulin signaling pathway is affected via PI3K (Brand et al., 2006), and the signal is regulated by FoxO in Drosophila (Jünger et al., 2003).
